Diazadispiroalkane derivatives - new viral entry inhibitors By Dr. Elke Prof. Dr. Bogner, Charité-Universitätsmedizin Berlin, Institute of Virology Abstract: Herpesviruses are widespread and can cause serious illness. Many currently available antiviral drugs have limited effects, result in rapid development of resistance and often exhibit dose-dependent toxicity. Especially for human cytomegalovirus (HCMV) new well-tolerated compounds with novel mechanisms of action are urgently needed. We investigated the antiviral effect of two small molecules (11826091 and 11826236) on HCMV infected human embryonic lung fibroblasts. Both substances were well tolerated and exhibited a strong dose dependent antiviral activity. Time of addition assays where 11826091 or 11826236 was added to cells before, before and during, during or after infection demonstrated an inhibitory effect on early steps Interestingly, 11826236 had an effect by addition to cells after infection. Detailed studies revealed th...
Maha Abdelgaber Abdellah Aly, Lecturer of Audiovestibular medicine, Faculty of Medicine, Assiut university, Egypt. Nonsyndromic hearing loss is an extremely heterogeneous disorder. Thus, clinical diagnostics is challenging, in particular due to differences in the etiology of hearing loss between populations. With this study, we wanted to elucidate the genetic basis of hearing loss in 61 consanguineous Egyptian families. In 25 families, linkage analysis was used as a prescreening to identify regions for targeted sequencing of candidate genes. Initially, the coding regions of 12 and later of 94 genes associated with hearing loss were enriched and subjected to massively parallel sequencing (MPS) with diagnostic yields of 36% and 75%, respectively. Causative variants were identified in 48 families (79%). They were found in 23 different genes with the majority being located in MYO15A (15.3%), SLC26A4 (9.7%), GJB2 (8.3%), and MYO7A (6.4%). As many as 32 variants were novel ones at the time ...
Shu Yun Heng, Singapore General Hospital "The acceptable incidence of thrombophlebitis following intravenous cannulation is 5%, as recommended by the Intravenous Nurses Society guidelines, but publications have reported startling figures of 20% to 80%. Given the frequency of intravenous lines, this presents a potential clinical problem. Thrombophlebitis can progress despite antibiotics to become a challenging clinical problem requiring surgical intervention. We aimed to determine the predisposing patient, catheter, and health care-related factors of peripheral vein thrombophlebitis in the upper extremity. We reviewed our experience of surgical intervention with analysis of the indications for intervention, description of the surgical procedures and outcomes. Qualitative syntheses revealed that patient-related factors that confer a higher risk included intercurrent illness, immunocompromised state, comorbidities such as diabetes mellitus, malignancy, previous thrombophlebitis, bu...
